The Long Amplicon Sequencing approach enables the sequencing of amplicon populations ranging from 600 bp to 15 kb that to the long reads technology of PacBio. It follows a two-step method:

Amplicon Generation: Amplification of a specific region with tags, performed in your laboratory. The amplicons can be generated either using unique tagged barcodes or through the Pacbio M13-tagged amplicons.
Library Preparation: Conducted using the PacBio protocol.

This initial amplification of the gene target is performed at your laboratory and it consists of the generation of long amplicons using one of the following approach

The PCR amplification targeting the region of interest is conducted using primers that include unique barcodes. This method generates amplicons through a single PCR step, significantly reducing associated biases.
